Is the MB service running under an admin id on that server? If yes then please check to see if there is a lock file in the backup folder and if there is delete it and try running it again.

It's running under an account that's a Power User.
Here is the backup location.
N:\Interwoven\MediaBin Server\MBDataDirectory\ImageDb\Backup\Full\MBDataBase.bak
I am about to check if the password being used by the backup is out of sync.
Here is a section of my MBBackup.Log

It was indeed a changed password for the username used for the odbc connection.
I performed the registry change indicated in this post and now the backup works properly.

This may be due to couple of reasons.
1) Check the user account has administrator previliges.
2) Check the Backup path is not remote IP; Better take back up using local path itself.
3) MBBackup.exe - execute the command line utility with local path. (This is equalent of taking back up using Enterprise manager)
