If we have a license for 40 concurrent users, what happens when the 41st person attempts to login? Do they get a message stating that the maximum number of connections has been exceeded? Do they get this message in the web client or desktop client?

They should be getting it on both the clients. In 4.6 when you reach such a state members of MBPAdmin group are still allowed to login. This is not so in 4.5.3.
