How Teamsite window Authentication will happens?

Can anyone pelase let me know how windows authentication will happens in TeamSite having an ACTIVE DIRECTORY as W2008R2 with Native Mode Group Usage.

We have configured the parameters in "iw.cfg", we want to know at what time the windows authentication will happens and at what point of time it fails to fetch the content in TeamSite.

Windows authentication occurs when you login
Authentication checks will happen if / when you invoke an operation and it is unable to find a valid cookie with your authentication
TeamSite doesn't fetch content
