Yes, It’s possible, please go to the properties page from the function menu of your folder then choose categories tab, there you should be able to find an option to create a category( provided you have enough permissions)

Yes, you can apply a category to a node using the UpdateNode() method and set the category info on the Node.Metadata property. As of update 10 there is also a SetNodeMetadata() method available.
